1. apply the isolator effect to all the samples in your beat and turn all knobs to the left so you hear no sound.
2. turn the effect off and start your loop and turn the effect off, while it's looping hold down effect grab button and press the isolator button and you will get a mute for as long as you hold the isolator button down.

It's similar to playing a sample pad with the gate mode on so you can rhythmically apply mutes to a resample and duck certain parts or sounds.
It's nice if you have a resample that is multiple generations and you want to remove a noise or sample that is overdubbed in your 2track like a snare etc,,

interesting, i woulda never thought of the isolator effect. what i normally did was just play the sample and then turn it all the way down as fast as i could so it would cut and then turn the volume up as fast as i could haha. nice to finally know how to actually do it. thank you! quick response too!

you can use other effects too, eg filter with cutoff turned all the way off... anything that ends up being silent

I prefer isolator because you can set different tones of it when you're not using it. So for one punch in, it would be silent. Next punch in, it would be be all audio but missing the low-end, etc.

But yeah, that punch-in with BPM+ISOLATOR = tits. :)
