So I have my 404, my scarlett 2i4 audio interface and my macbook pro... I have my RCAs running from my line out on my 404 into the lines in the back of the interface, which is connected to laptop via usb.

I tested the Roland SP-606 for audio-out a while back and it works with no issues. I can't remember if I tested the USB sample functionality, but I'm 99% sure that works.

It sounds like the SP-555 would be the most-excellent addition to your set-up.
The loop pedal that you posted (Ditto Looper) is MONO, which isn't a problem if you sample in MONO. Be aware, I don't think you can seamlessly transition from one loop to another without stopping playback on the DItto ...

What loop pedal are you looking to buy?
You'll want to look at impedance, but I'm assuming that RCA/unbalanced (like most of the SP series) will work fine using a rca to 1/4" cable to a loop guitar pedal (as most seem to be in stereo).
